The government of Pakistan owns the site, and keeps changing its mind on whether or not to sell and redevelop. You could build a huge tower on that site, even without the Midtown East upzoning.
That site has been host to many proposals over the years. One day, something very tall and prominent will rise on that site. It would have been a good site for Nordstrom, though the site they eventually chose seems pretty good too.

Yes, it's in the hot zone, and can be built as large as any of the towers around Grand Central, with some "starchitecture". Look for more on that in the future.
What I find interesting about that store plan is that it's basically the same as is planned for this site...
