This is basically the "Niagara-GTA Corridor" studied a little while back by MTO, often referred to as the
mid-peninsula highway.
There are tricky parts (e.g., the highway alignment around Ancaster, where to put the crossing of the Welland Canal) but the general idea is the same.

Could also widen/improve the 403 and Hwy 6 South before building a connector through the Niagara Peninsula, though the 403 has limitations, and expensive work-arounds for them.
